Separation of Biomolecules such as Antibody Drug Conjugates (ADCs) and Monoclonal Antibodies (mAbs) Using Various Analytical Chromatographic Modes and Factors Affecting it
Abstract: ADCs are more complex and heterogeneous than the corresponding antibodies. Different chromatographic modes are selected for both large scale and analytical scale of purification, based on the specificity of the individual ADC, the nature of the linker and the attachment sites etc. With the advancement of HPLC analytical column chromatography the separation of complex peptide mixtures derived from protease digestions of mAbs and ADCs are possible. Here we present a brief overview of the separation of monoclonal antibodies and ADC using different modes of analytical chromatographic techniques.
Atis Chakrabarti earned a Ph.D. in Chemistry from North Bengal University, India on the design and synthesis of anti-proliferative probes. His post-doctoral research at the Indian Institute of Science (IISc) Bangalore, India focused on the molecular biology and biophysical characterization of bacterial proteins and stability studies. In 2000, he joined Kimmel Cancer Center at Thomas Jefferson Medical College and University in Philadelphia, researching diagnostic and therapeutic probes for PET imaging while utilizing different types of chromatography for the purification of the probes, as well as on transposon based gene therapy. He is an ex-adjunct faculty member of Scranton University, PA where he taught toxicology to the MS students. He is also an ex-adjunct faculty member in the department of chemistry at Camden County College, NJ, teaching Organic Chemistry.
Atis joined Tosoh Bioscience in July 2008 as a Technical Specialist. In October 2011, he became the Manager of Technical Service for the Laboratory Products division. Currently, and since August 2013, he is working as the Manager of Technical Service, covering the technical service of analytical chromatography columns and process chromatography resins. He leads a talented group of technical service specialists. His group is involved in developing applications on analytical chromatography TSKgel columns and process media TOYOPEARL and TSKgel resins. His technical service group supports customers working on the isolation and purification of biopolymers using all modes of chromatography. Atis' research is focused on expanding the range of Tosoh products in separation science.
